Background

p300 is a 2,414-residue human transcriptional coactivator that contains multiple conserved domains—bromodomain, CH1–CH3, HAT, KIX, and ZZ-type zinc finger—that enable it to serve as a histone acetyltransferase and a scaffold bridging sequence-specific DNA-binding factors with chromatin-remodeling machinery; by acetylating histones H3 and H4 as well as non-histone substrates such as p53 and NF-κB, it opens local chromatin structure and potentiates transcription, while its own activity is tightly regulated through phosphorylation, auto-acetylation, and competitive binding with viral oncoproteins like E1A or cellular partners like CBP; because it integrates proliferative, differentiation, and DNA-damage signals, p300 is indispensable during embryogenesis and is frequently altered—mutated, deleted, or translocated—in cancers, making it both a tumor suppressor and, in certain contexts, an oncogenic driver, and its HAT activity is pharmacologically targeted by small-molecule inhibitors under investigation for cancer therapy.
