Background
The Androgen Receptor splice variant 7 (AR-V7) is a constitutively active, ligand-binding domain–deleted form of the androgen receptor that retains the N-terminal transactivation domain and DNA-binding domain, enabling ligand-independent nuclear translocation and transcriptional activation of AR target genes; its expression, driven by alternative splicing or gene rearrangements, is markedly enriched in metastatic castration-resistant prostate cancer and functions as a key driver of resistance to second-generation anti-androgens such as enzalutamide and abiraterone by sustaining AR signaling even under systemic androgen suppression or pharmacologic AR inhibition.
