Net als in eerdere Devil May Cry games moet je het ook in dit vierde deel opnemen tegen hordes demonen, duivels en andere sinistere figuren. Gewapend met een zwaard, verschillende geweren, magische spreuken en een grote bek moet je afrekenen met deze schurken om de mensheid wederom te redden. Het grote verschil met eerdere delen is dat je niet met Dante, maar met Nero speelt.

Net als Dante is Nero bewapend met een zwaard en een gun, maar heeft daarnaast ook een zogenaamde Devil Hand, waarmee hij vijanden naar zich toe kan trekken. Dit met als enig doel om hem vervolgens genadeloos af te maken. Ook is dit handig om de combo waar je mee bezig bent door te laten gaan.

Naast de harde actie beschikt Nero ook over magie, waarmee hij speciale aanvallen kan inzetten. Denk daarbij aan het omhoog rammen van een vijand om hem vervolgens in de lucht te killen of met een enorme sprong in een keer op diezelfde vijand te springen. Er lijken weer genoeg mogelijkheden aanwezig te zijn om al je agressie van je af te spelen.
